You want to specify lpi in AI?
You need to be printing to a postscript printer, such as the one included in AI.
Change the print output settings to separations and choose your lpi.

What is it exactly that you need to determine the LPI for? Unless you're using halftones, lineart done in Illustrator should be printed at more than sufficient resolutions for screen printing purposes. Just stay away from lines under 1-2pt until you get comfortable with the process!

Hope this helps!
